Maintenance of liquid chromatography
1. The daily operation of HPLC conditions:
Temperature :10-30 ℃;
Relative humidity "80%;
Best constant temperature and humidity, away from the high electrical interference, high vibration equipment .
2. Pump maintenance:
1) the use of mobile phase to clean as much as possible;
2) into the liquid at the head to the sand core filter regular cleaning;
3) the mobile phase to prevent sediment exchange;
4) to avoid clogging the pump or air bubbles;
3. Injector maintenance:
1) After the end of each analysis, rinsed repeatedly injector, prevent cross contamination between analysis instruments,
4. Column maintenance:
1) The column can not impact in any case, bending or strong vibrations;
2) When connected chromatography columns, and when the valve or piping must be cleaned;
3) pay attention to the mobile phase degassing;
4) Avoid the use of high viscosity of the solvent as mobile phase;
5) samples to be purified sample;
6) strict control of sample volume;
7) After a day of work, to clean the injection valve in the remaining samples;
a day of determination of the end, should be cleaned with appropriate solvent column;
9) If the analytical column for long periods of use, application of appropriate organic solvent to save and closed;
5.Detection device (UV) Maintenance:
1) UV lamp maintenance: In the analysis, the column was almost balanced, open the detector; in the analysis is complete, immediately turn off the detector.
2) the sample cell to be maintained.
